The Horton Lab will measure the function of HIV-epitope-specific T cells identified in this project: "Tregs Differentially Suppress HIV-specific CD8+ T Cells" and this project: "Identifying Characterizing and Inducing Effective Anti-HIV T Cell Responses." These assays will use intracellular cytokine staining (ICS) assays to measure IFN-γ, TNF-α, IL-2 and Granzyme B in response to HIV-1 epitopes. In addition we will perform Carboxy-fluorescein diacetate, succinimidyl ester (CFSE) assays to measure proliferative ability of these HIV-specific T cells.
